Materials:
Bacterial culture 24-48 hrs old, 3-5% H2O2
Procedure:
A. On a plate/slant
1. To a plate or slant add 2 drops of H2O2
2. Observe for the evolution of
bubbles (catalase positive reaction)
B. On a slide
1. Pick a colony from a plate
with a wooden applicator stick (do not use a loop)
2. Smear colony on a clean, dry
slide and apply 5 drops of H2O2
3. Observe for the evolution of
bubbles (catalase positive reaction)

A. On a plate/slant
1. To a plate or slant add 1 ml
of Benzidine reagent
2. Now add 1 ml of 3-5% H2O2
3. Observe for the development
of a blue color (positive porphyrin test)
B. On filter paper
1. Pick a colony from a plate with
a wooden applicator stick (do not use loop)
2. Add 1 ml Benzidine reagent
3. Add 1 ml 3-5% H2O2
4. Observe for the development
of a blue color (positive prophyrin test)
